Edit the Lemmings 2 profile in D-Fend, go to the General tab and turn on "Auto lock mouse". That should fix it -- click in the DOSBox window when it's running to give the game mouse control, and press the Win key if you need to get the mouse back into Windows.

What spec machine are you playing on? Increasing the number of cycles the game is getting can help performance, but only if you have a machine that can cope.

else look for the sensitivity option.
it's 100 by default but maybe 200 is better for you
